The easiest way to accomplish all objectives is to take the challenge twice. The first time, equip powerful Magic medals and open every chest, making sure to slay the enemy Pretenders in one turn. The second time, simply go straight to the target enemy and slay it, avoiding all treasure chests.
In order to accomplish all objectives in one go is to first open the item chest located on the stairs in the bottom-left, which contains a Great Pretender. After defeating the enemy, head straight to the target without opening any other chest.
